cilinder problem need HELP!!

Hi guys.....

I jus fixed up my k10 carb and and i noticed one of my cilinders arnt workin... when i removed one of the plug weires the engine dose not make any difference... i checked the plug, plug weire etc.. and evn checked the tappets and everything is fine... pls tell me wot could be wrong.....
